Dopis o toleranci je nejdůležitější ze čtyř listů, které slavný anglický myslitel 17. století John Locke věnoval tématu, jež až dodneška zaujímá politiky, filosofy i laické čtenáře. Je odpovědí na projevy náboženské a politické nesnášenlivosti doby, v níž vznikl (1689), neztratil však nic ze své naléhavosti. Locke v tomto díle spojil empirický pohled na lidskou společnost s vírou v existenci přirozeného zákona, který Bůh vložil v řád světa, aby umožnil lidem žít v rozumném společenství. Takové společenství je však třeba prakticky zabezpečit. Locke přichází s ohromující myšlenkou: je nutné přísně oddělit veřejný zájem od zájmu soukromého. Víra a různé formy náboženství, jakož i jejich praktické projevy v podobě jednotlivých církví a sekt, představují zájem soukromý, do něhož nesmí stát nijak zasahovat. Církve zase nesmí pod žádnou záminkou zasahovat do záležitostí veřejných.

Svou třetí částí vytváří novověkou sémantiku. V závěrečné čtvrté části formuluje stupně a meze poznání, včetně toho, jak poznávat „deisticky“ Boha. Dílo vyprovokovalo Leibnize k obšírné replice a Berkeleye k psychofyzickému paralelismu. Kniha patří mezi základní publikace novověkého myšlení. Autor je považován nejen za klasika tzv. empirismu, ale právem se řadí také mezi stěžejní autory moderního liberalismu.
