Homunculus erzählt die fiktive Geschichte des Mediziners Manabu Ito und seines "Versuchskaninchens" Susumu Nakoshi - die Geschichte einer so genannten Trepanation. In der Neurochirurgie bezeichnet man mit Trepanation die operative Öffnung des Schädels zur Vornahme von Eingriffen im Schädelinnern oder auch zur Senkung des Hirndrucks. Ito will erforschen, welchen Einfluss eine solche Schädelöffnung auf das menschliche Bewusstsein hat. Nakoshi willigt in die OP ein. Danach ist nichts wie bisher - Nakoshis Wahrnehmung hat sich grundlegend verändert. Sobald er ein Auge schließt, sieht Nakoshi z.B. Robotermenschen, Mädchen aus Sand, Frauen ohne Hals, kurz: deformierte Körper - überall. Er erkennt das "wahre Ich" eines jeden Menschen, den Kern ihres Wesens, das Innerste nach außen gekehrt. Was genau sich der Wissenschaftler Manabu von dem Experiment erhofft, bleibt noch undurchsichtig. Ebenso die Frage, was Nakoshi wohl sieht, wenn er sich selbst betrachtet? Macht Euch auf eine furiose Entwicklung des Plots in Band 4 und 5 gefasst!
Im Dienste der Wissenschaft Gibt es ihn vielleicht doch, den Homunculus, den kleinen Mann im Kopf eines jeden Menschen, der - so glaubte man früher - die Geschicke und Gefühle steuert? Die Inhalte des Medizinstudiums reichen nicht aus, um Manabu Itos Wissensdurst zu stillen. Er will das menschliche Gehirn erforschen und zwar den Teil, der bislang noch nicht mit medizinischen und biochemischen Erkenntnissen ergründet werden konnte. Susumu Nakoshi weiß indes nicht so recht, welchen Effekt es haben wird, wenn Manabu ihm ein Loch in den Schädel bohrt. Gesund klingt es nicht, aber dann braucht Susumu das Geld für sein geliebtes Auto und 700.000 Yen scheint ihm ein guter Preis zu sein für die Einwilligung in das Experiment, das leider nicht so verläuft, wie Susumu es sich wünscht. Denn mit dem Eingriff beginnt sein Leben aus den Fugen zu geraten. Hideo Yamamoto forschte mit seinem Yakuza-Epos "Koroshiya 1", auch bekannt unter dem Titel der Verfilmung "Ichi - The Killer", nach Ursache und Wirkung von Gewalt und trieb deren Darstellung auf die Spitze. Mit "Homunculus" führt Yamamoto seine Studien des menschlichen Verhaltens mit psychologischem Gespür für seine Figuren ohne die Schockeffekte schonungslos präsentierter Metzeleien weiter.
