Nejznámějším Suetoniovým dílem jsou bezesporu jeho Životopisy císařů (De vita Caesarum) o osmi knihách. Tuto knihu napsal Suetonius na vrcholu své úřednické dráhy v letech 115 - 120 n.l. a věnoval ji prefektu pretoriánské gardy Septiciovi Clarovi. V této knize popisuje Suetonius císaře počínaje Caesarem jakožto zakladatelem dynastie a konče Domitianem. Jsou nabity konkrétními fakty a údaji o nejintimnějších podrobnostech z jejich života. Pro velké množství detailů není látka uspořádána chronologicky, nýbrž podle určitého schematu, zahrnujícího rozličné stránky státnické činnosti i soukromého života zobrazované osoby. Tato forma Suetoniovy práce bývá často kritizována pro svoji úroveň "bulvárního plátku", ale zůstává faktem, že Životopisy císařů byly a jsou velmi úspěšným dílem, které přiblížilo božské císaře prostému lidu. Našlo si i velké množství pokračovatelů, jeho dílo se totiž stalo jakýmsi vzorem pro autory biografií minimálně do středověku.
