Focusing on innovative drug delivery, this work presents an amphoteric liposomal composition designed for effective oligonucleotide transport both in vitro and in vivo. These liposomes feature pH-sensitive lipids that adapt their charge based on environmental pH, enhancing nucleic acid stability under acidic conditions while preventing aggregation with serum components at physiological pH. Their optimized design facilitates pH-dependent fusion with endosomal membranes, addressing key challenges in oligonucleotide delivery and maximizing therapeutic potential.
